The Bet365 in human cells is very complex, lengthAbout 2 meters of DNA is entangled in the micron-level nucleus,Transcription is performed in an orderly manner in a highly complex structure、Copy、Repair and other functions,Regulate complex gene expression system。Single cell data display,Bet365 changes dynamically over time,And there may also be heterogeneity in the Bet365 of the same type of cells。Then,How to characterize the changes in Bet365 that can affect gene expression or even cause disease?Around this issue,We start from the basic unit of Bet365—chromatin topologically associated domain,TAD)Get it,Characterize Bet365、Quantitative characterization,And mine TAD-related tumor markers: 1) Constructed the chromatin open density index SDOC to characterize the activity of TAD;2) Based on TAD boundary changes and transcription abnormalities,Constructed the TARGET computational framework to screen oncogenes;3) Changes based on TAD hierarchical structure,Definition of TH score indicator,It was found that changes in TH score of genes are significantly related to the malignant phenotype and poor prognosis of tumors。
